Abstract

Research has been carried out on modification of tire tuber starch (Amor- phophallus onchopyllus) bypregelatination and acetylation as a disintegrant in paracetamol tablets with wet granulation method. The purpose of this study was to determine whether the modified tire tuber starch by pregelatination and acety- lation can produce starch with good characteristics, and to determine the good tire tuber starch concentration as a tablet crushing agent that can produce quality tablet preparations that meet the requirements, and also to determine the quality tablet preparation with modified tire starch by pregelatination and acetylation. This study used a laboratory experimental method with the use of two starch mod- ification methods and six tablet formulas with three starch concentrations (5%, 10%, and 15%) in each starch modification method. Evaluation of modified starch and granule formula includes (moisture test, flow property test, angle of repose test, and Carr index test). The tablets were evaluated including (weight uniformity test, tablet hardness test, friability test, disintegration time test, and dissolution test. The results of the research on modified starch and granule formulas have a moisture content value of 3-5%, flow properties <10 g/s, angle of repose 20- 40o, and Carr Index 12-18%. In tablets, all formulas had uniform weight, tablet hardness 4-5 Kg, tablet friability <1%, disintegration time did not exceed 15 minutes, and dissolution level of paracetamol was >80%.
